چکیده

During the past five years ganglion-blocking agents have been used extensively in the treatment of arterial hypertension. The discovery of the activity of hexamethonium by Paton and Zaimis (1949) placed at our disposal an active therapeutic agent capable of producing a substantial reduction in the blood pressure. Subsequent research has been directed to the synthesis of new drugs offering advantages over hexamethonium. In the treatment of hypertension, it is desirable to obtain a sustained fall in the blood pressure by the oral administration of a drug producing a minimum of sideeffects. Hexamethonium is inconstant in action by mouth, its duration of action is short, and it produces side-effects characteristic of generalized ganglionic blockade. Maxwell and Campbell (1953) examined a number of methonium homologues and indicated the suitability of one of them, pentolinium, for therapeutic application. At present pentolinium is the most active hypotensive drug available and it has been widely employed. Trapold et al. (1955) have described the pharmacological actions of a new synthetic compound. " ecolid " (SU-3088), and Grimson et al. (1955) have demonstrated its activity in lowering the blood pressure in man. Trapold et al. (1955) have described a series of halogenated isoindolines with potent ganglion-blocking activity of long duration. Ecolid was the most active compound investigated. It is 4,5,6,7-tetrachloro-2-(2-dimethyl-aminoethyl)isoindoline dimethochloride, the structural formula being as follows: Cl CH3

Deliberately Lowering Blood Pressure with Sodium Nitroprusside

The goal of hyptensive anesthesia is the safe reduction of intraoperative blood loss, thus reducing transfusion risks.  Previous hypotensive technics had a narrow margin of safety in that cardiac output was usually reduced in proportion to the degree of induced hypotension. This is in contrast to the results of our study, in which Sodium Nitroprusside was used as the sole hypotensive agent. Ni...

Rho Kinase Inhibitors as a Novel Treatment for Glaucoma and Ocular Hypertension

In an elegant example of bench-to-bedside research, a hypothesis that cells in the outflow pathway actively regulate conventional outflow resistance was proposed in the 1990s and systematically pursued, exposing novel cellular and molecular mechanisms of intraocular pressure (IOP) regulation. The critical discovery that pharmacologic manipulation of the cytoskeleton of outflow pathway cells dec...

Effects of Norepinephrine on the Coronary Circulation in Man.

T HE sympathomimetic amine l-norepinephrine is extensively used in the treatment of hypotensive states, and is considered by many to be the pressor agent of choice.1-3 Shock associated with myocardial infarction is one of the more important of these hypotensive states.4-8 Therefore, it seemed relevant to examine the effects of infusion of this agent upon the coronary circulation in the human su...

Responses of the ballistocardiogram in hypertensive patients to the acute administration of apresoline, hexamethonium, veratrone, regitine and sodium amytal.

Acute ballistocardiographic responses to five hypotensive drugs have been evaluated in 15 patients with essential hypertension in conjunction with depressor effects. In the majority of experiments qualitative and quantitative ballistocardiographic improvement paralleled the pressure reductions. On occasion the reverse was true. Inferences were drawn from these reactions based on the pharmacodyn...

Long-term treatment of severe hypertension with minoxidil.

Minoxidil, a new potent hypotensive agent, was used as the primary antihypertensive agent in 11 patients--10 men and 1 woman aged 35 to 54 years with severe hypertension that was refractory to treatment with maximal (or maximally tolerated) doses of conventional antihypertensive agents. Six patients had severely impaired renal function and three of them were undergoing long-term hemodialysis. T...
